Nitric oxide (NO) is widely distributed in the nervous, circulatory, respiratory, digestive, and urogenital systems of organisms, and is particularly abundant in neural tissues. NO is a novel biological signaling molecule that mediates intercellular and intracellular signal transduction. Due to its free radical nature, NO is readily oxidized to nitrite (NO₂⁻) and nitrate (NO₃⁻). Therefore, total NO content is generally estimated by measuring the sum of NO₂⁻ and NO₃⁻. This kit employs a modified Griess method for detection. In this method, nitrate (NO₃⁻) is first reduced to nitrite (NO₂⁻). Under acidic conditions, NO₂⁻ reacts with sulfanilamide to form a diazonium salt via diazotization, which then couples with N‑(1‑naphthyl)ethylenediamine dihydrochloride to produce a purple‑red complex. The absorbance of this complex is measured at 540 nm, allowing for the calculation of NO content in the sample.
