Sorbitol is the primary translocated sugar in Rosaceae plants such as apple and pear. After being transported to the fruit, it is converted into other sugars. Sorbitol oxidase (SOX) is one of the key enzymes involved in this conversion process, catalyzing the oxidation of sorbitol to glucose, and is closely associated with fruit quality formation and sugar accumulation. This kit employs the DNS colorimetric method for the determination of SOX activity. The principle is as follows: SOX catalyzes the conversion of sorbitol to glucose. Under alkaline conditions, glucose undergoes an oxidation‑reduction reaction with 3,5‑dinitrosalicylic acid (DNS) to generate brown‑red 3‑amino‑5‑nitrosalicylic acid, which exhibits a characteristic absorption peak at 540 nm. Within the established linear range, the rate of increase in absorbance at 540 nm is directly proportional to the SOX activity in the sample, and the enzyme activity can be quantitatively calculated using a standard curve.
