Carbohydrates are one of the important components of plant tissues, serving as both primary metabolites and storage substances. Total sugar (i.e., total carbohydrates) includes soluble monosaccharides, disaccharides, and insoluble polysaccharides such as starch, cellulose, and chitin. This kit employs an acid hydrolysis‑DNS colorimetric method for the determination of total sugar content: Total sugars in the sample are hydrolyzed to reducing sugars under acidic conditions. In the presence of NaOH and glycerol, the reducing sugars react with 3,5‑dinitrosalicylic acid (DNS) upon heating, reducing DNS to an amino compound, which appears orange‑red in excess alkaline solution. This product exhibits a maximum absorption peak at 540 nm. By measuring the absorbance at 540 nm, the total sugar content in the sample can be quantitatively calculated.
