The degree of pectin methylesterification (PMD) determines the number of negative charges and is closely related to the adsorption of heavy metals by plant root cell walls and the resistance to heavy metal toxicity. The pectin content of the cell wall and its degree of methylesterification directly affect the plant's ability to withstand heavy metal toxicity. In the assay, pectin methyl ester bonds are released by saponification to generate methanol, which is oxidized by alcohol oxidase to formaldehyde. Formaldehyde then reacts with 2,4-pentanedione to produce a colored product with a characteristic absorbance peak at 412 nm. Meanwhile, galacturonic acid undergoes a condensation reaction with carbazole in a sulfuric acid solution to form a purple-red product with a characteristic absorbance peak at 530 nm. The degree of pectin methylesterification (PMD) is calculated as the ratio of methanol production to galacturonic acid content.
