Product Introduction

Polyvinyl Alcohol-Glycerol Mounting Medium is a type of Fluorescence Decay-Resistant Mounting Medium, a mounting agent designed to slow down fluorescence quenching. It can reduce the quenching effect of various common fluorescent dyes, with its anti-quenching performance significantly superior to that of regular anti-quenching mounting media. Generally, the anti-quenching effect only starts to diminish after 1–2 weeks. Based on a dosage of 50-100 μl per sample, 5 ml of the reagent can be used for mounting at least 50 samples. This reagent is intended for research purposes only and not for clinical diagnosis or other applications.

Operating Procedures (For Reference Only)

1. Adherent Cell Samples

① After staining, aspirate the staining solution completely.

② Add 1 drop of Polyvinyl Alcohol-Glycerol Mounting Medium onto the slide, then place the coverslip with adherent cells onto the medium. Ensure the cells are in contact with the mounting medium and minimize bubble formation.

③ Observe under a fluorescence microscope immediately, or store the slides at 4 °C away from light for 1-2 weeks.

2. Tissue Sections

① After staining, aspirate the staining solution completely.

② Add 1 drop of Polyvinyl Alcohol-Glycerol Mounting Medium onto the tissue section, then place a coverslip over it. Ensure the section is in contact with the mounting medium and minimize bubble formation.

③ Observe under a fluorescence microscope immediately, or store the slides at 4 °C away from light for 1-2 weeks.

3. Other Samples

Follow the procedures for adherent cell samples or tissue sections.

Precautions

1. The concept of anti-fluorescence quenching is relative. Stained samples are best observed immediately after staining; if immediate observation is not feasible, observation should be completed on the same day.

2. All fluorescent substances are prone to quenching. Stained samples should be stored at 4 °C away from light.

3. Although this Polyvinyl Alcohol-Glycerol Mounting Medium can slow down quenching, samples should still be protected from light as much as possible during use and storage.

4. Antibodies labeled with rhodamine cannot be mounted with this medium; Polyvinyl Alcohol Fluorescent Mounting Medium is recommended instead.

5. For your safety and health, wear a lab coat and disposable gloves during operation.

6. Use the reagent as soon as possible after opening to avoid compromising subsequent experimental results.
