Overview

Xylanase (EC 3.2.1.8) is mainly produced by microorganisms and catalyzes the hydrolysis of xylan. It is also known as pentosanase or hemicellulase. This enzyme can decompose cell walls and β‑glucan in raw materials used in the brewing and feed industries, thereby reducing the viscosity of the mash during brewing and promoting the release of active substances. Meanwhile, it reduces non‑starch polysaccharides in feed, facilitating nutrient absorption and utilization. Consequently, it is widely applied in both the brewing and feed industries. Alkaline xylanase (BAX) is generally isolated from microorganisms with an optimal growth pH of 9–11. The assay principle of this kit is as follows: BAX catalyzes the degradation of xylan into reducing oligosaccharides and monosaccharides under alkaline conditions. The resulting reducing sugars undergo a chromogenic reaction with 3,5‑dinitrosalicylic acid (DNS) in a boiling water bath, generating a colored product with a characteristic absorption peak at 540 nm. The color intensity of the reaction solution is directly proportional to the amount of reducing sugars produced by enzymatic hydrolysis. Therefore, the BAX activity can be calculated by measuring the rate of increase in absorbance at 540 nm.
