Overview

Cerebrospinal Fluid (CSF) is a colorless and transparent fluid found in the cerebral ventricles, subarachnoid space and central canal of the spinal cord. It is secreted by the choroid plexus within the ventricles and bears similar properties to blood plasma and lymph. In healthy adults, the total volume of CSF is approximately 100~150 mL. It is weakly alkaline and free of red blood cells. Containing specific chemical components and pressure, CSF plays a vital role in maintaining stable intracranial pressure. When the central nervous system is damaged, CSF examination serves as a crucial auxiliary diagnostic method.Pandy’s Reagent is mainly composed of phenol. The globulins in cerebrospinal fluid bind with phenol to form insoluble protein salts that precipitate, resulting in white turbidity or sediment. This assay is also known as the Pandy test, which is widely applied for qualitative detection of protein in human or animal cerebrospinal fluid. This reagent is intended for research use only and shall not be used for clinical diagnosis or other purposes.

Operating Procedures (For Reference Only):

1. Take a small test tube and add 2~3 mL of Pandy’s Reagent.

2. Add 1~2 drops (approximately 0.05~0.1 mL) of cerebrospinal fluid, mix thoroughly, and observe immediately against a black background.

Result Interpretation:

ResultObservation
NegativeClear and transparent, no opalescence
Weakly positive (±)Slight white opalescence, only visible against a black background
Positive (+)Grayish white opalescence
(2+)White turbidity
(3+)Thick white flocculent precipitate
(4+)White clot

Precautions:

1. The sample shall be sent for test immediately after collection, and the waiting time generally shall not exceed 1 hour.

2. The result is mostly negative or weakly positive under normal conditions; positive reactions will appear when lesions occur in brain tissue or cerebrospinal cord.

3. Please use up the reagent as soon as possible after opening to avoid adverse effects on subsequent experiments.
