Overview

Escherichia coli (E. coli) is a Gram-negative, short rod-shaped bacterium, approximately 0.5 µm × (1–3 µm) in size. It is peritrichously flagellated, motile, non-spore-forming, and capable of fermenting various sugars to produce acid and gas. It is a normal commensal inhabitant of the intestinal tract of humans and animals. After birth, E. coli colonizes the infant gut through feeding and remains throughout life, accounting for approximately one-third of the fecal dry weight. DNA obtained from E. coli is of relatively high yield but moderate purity, which is generally sufficient for most molecular biology experiments. CTAB (cetyltrimethylammonium bromide) is a cationic detergent. In solutions of high ionic strength, CTAB forms complexes with proteins and polysaccharides (with the exception of most acidic polysaccharides), but does not bind to nucleic acids. By adjusting the salt concentration, nucleic acids can be released from CTAB complexes and precipitated, enabling DNA purification from polysaccharide-rich organisms (such as plants and certain Gram-negative bacteria). This product is a convenient CTAB-based genomic DNA extraction kit for E. coli. The principle is as follows: proteins are digested by proteinase K; polysaccharides are precipitated by CTAB; proteins are denatured and separated from DNA by heating; and finally, genomic DNA is obtained by ethanol precipitation. The resulting DNA can be used in downstream applications such as restriction enzyme digestion and PCR. This kit is intended for research use only and is not suitable for clinical diagnosis or other applications.
