Serum total iron‑binding capacity (TIBC) refers to the ability of serum transferrin to bind iron. Its level is closely associated with the occurrence of diseases such as iron deficiency anemia and acute hepatitis. This product provides a simple, sensitive, and rapid method for TIBC determination, suitable for serum or plasma samples. The detection principle is as follows: Fe²⁺ reacts with ferrozine to form a purple‑red compound, which exhibits a characteristic absorption peak at 562 nm. Under alkaline conditions, serum transferrin binds to Fe³⁺; the remaining unbound Fe³⁺ is reduced to Fe²⁺, and the measured absorbance A₁ is proportional to the amount of unbound Fe³⁺. After acidification treatment, the transferrin‑bound Fe³⁺ is released and further reduced to Fe²⁺, and the measured absorbance A₂ is proportional to the amount of total Fe³⁺. The difference between A₂ and A₁ (A₂ − A₁) is directly proportional to the TIBC concentration.
