Soil cellulase (S-CL) is mainly derived from soil microorganisms. It catalyzes the degradation of cellulose to produce glucose and acts as one of the key enzymes involved in soil carbon cycling. Glucose generated from the enzymatic breakdown of crop straw serves as an important carbon nutrient source for the growth and metabolism of soil microbes. This kit uses the anthrone colorimetric method to measure S-CL activity: S-CL catalyzes the hydrolysis of cellulose (or sodium carboxymethyl cellulose) to yield reducing sugars. Under concentrated sulfuric acid, reducing sugars undergo dehydration to form furan compounds, which react with anthrone reagent to produce a blue-green product with a characteristic absorption peak at 620 nm. The enzymatic activity of S-CL can be quantitatively calculated by measuring the increasing rate of absorbance at 620 nm.
