Overview
The quantitative determination of β-glucan is based on a series of coupled enzymatic reactions: first, β-glucan is hydrolyzed by lichenase to oligosaccharides, which are then completely degraded to glucose by β-glucosidase. Glucose is oxidized by glucose oxidase to gluconic acid, with concomitant production of H₂O₂. Subsequently, in the presence of peroxidase, H₂O₂ reacts with 4-aminoantipyrine and phenolic derivatives to form a purple compound, which exhibits a maximum absorbance peak at 540 nm. Therefore, by measuring the change in absorbance of the reaction system at 540 nm, the β-glucan content can be accurately determined.
