Overview
Trypan blue is a cell‑impermeable dye that cannot penetrate the intact plasma membrane of living cells. Therefore, viable cells remain unstained or show only a very faint background color. When cells are damaged or dead, the integrity of the plasma membrane is lost, allowing trypan blue to enter the cells and bind to macromolecules such as proteins, thereby staining the dead cells a distinct blue color. By observing the extent and intensity of blue staining, the distribution and relative proportion of dead cells in the tissue can be determined.
