Overview
ATP-Citrate Lyase (EC 4.1.3.8) is a key enzyme in fatty acid synthesis. The acetyl-CoA it generates can be utilized for fatty acid synthesis and chain elongation, flavonoid synthesis, amino acid synthesis, and other metabolic processes.
Assay Principle: In the presence of ATP and coenzyme A, ACL catalyzes the cleavage of citrate into acetyl-CoA, oxaloacetate, ADP, and inorganic phosphate. Malate dehydrogenase further catalyzes the conversion of oxaloacetate and NADH to malate and NAD⁺. The activity of ACL is calculated by measuring the rate of decrease in NADH absorbance at 340 nm.
