Overview

β-1,4-Galactosyltransferase (LgtB), EC 2.4.1.90, derived from Neisseria meningitidis.This enzyme transfers galactose via a β‑1,4 linkage to oligosaccharide chains with non‑reducing‑terminal GlcNAc or glucose. It is currently the most efficient enzyme for enzymatic synthesis of LacNAc. Recombinantly expressed in Escherichia coli, this product exhibits high purity and high activity.

Reaction Equation

GlcNAc + UDP‑Gal → LacNAc + UDP

Usage Instructions

  1. Take out the enzyme and dissolve it in an appropriate amount of purified water, mix well and set aside. Do not repeatedly freeze‑thaw the dissolved enzyme; store it at 2–8 °C.

  2. Add substrates and sugar nucleotide donor (UDP‑Gal), 5 mM MnCl₂ to the reaction system. Appropriate buffer salts (e.g., Tris‑HCl) may be supplemented to adjust pH to 6.0, then add an appropriate amount of LgtB according to reaction requirements. Alternatively, UDP‑Gal can be synthesized via a one‑pot multi‑enzyme method by adding enzymes GalK and AtUSP, substrates ATP, UTP, and acceptor substrates. Adjust pH to 6.0 and add an appropriate amount of LgtB according to reaction requirements.

  3. After mixing, incubate the sample at 37 °C for 1–4 hours.

Note: When scaling up the reaction system, adjust incubation time or enzyme dosage according to actual conditions; low‑speed shaking can be used for homogenization.
