Overview
β-Glucuronidase (β-GUS, EC 3.2.1.31) is a widely distributed hydrolase found in animals and microorganisms; however, endogenous GUS activity is absent in the vast majority of plant cells and many bacteria and fungi. Consequently, the GUS gene is extensively used as a reporter gene in transgenic plants, bacteria, and fungi. This kit employs p-nitrophenyl-β-D-glucuronide as the substrate. β-GUS hydrolyzes the substrate to generate p-nitrophenol (PNP), which exhibits a characteristic absorbance peak at 405 nm. By continuously monitoring the initial linear increase in OD₄₀₅ per unit time (ΔOD₄₀₅/min) in kinetic mode, the β-GUS enzyme activity in the sample can be precisely calculated.
