Overview
Carboxylesterase (CarE) in mammals, also known as aliesterase, is widely distributed in tissues and organs and belongs to the serine hydrolase family. CarE catalyzes the hydrolysis of endogenous and exogenous substances containing ester bonds, amide bonds, and thioester bonds, but it does not catalyze the hydrolysis of acetylcholine or its analogs. CarE is involved in lipid transport and metabolism and is associated with the detoxification and metabolism of various drugs, environmental toxins, and carcinogens. Organophosphate pesticides can bind to and inhibit CarE activity.
Assay Principle: CarE catalyzes the hydrolysis of 1-naphthyl acetate to produce naphthol, which subsequently reacts with a Fast Blue dye to form a colored product. The increase in absorbance at 450 nm is measured, and the rate of increase is used to calculate CarE activity.
