This buffer contains highly purified casein molecules and proprietary blocking factors, delivering superior blocking performance. It is designed for saturating blocking of blotting membranes and other solid-phase surfaces, effectively reducing background signals, lowering intra-assay and inter-assay coefficients of variation (CV), and enhancing detection sensitivity. The product is also suitable for blocking other protein-coated surfaces and compatible with most detection systems.
Basic Product Information
• Buffer: Tris
• pH: 7.2 ± 0.2
• Protein: Highly purified casein
• Preservative: 0.0015% (w/v) Isothiazolinone
• Shelf life: 2 years
• Storage temperature: 2–8°C
• Application: Western blot (Dilute to 1× working solution before use; suitable for blocking blotting membranes, diluting primary antibodies and enzyme-conjugated secondary antibodies.)
Instructions for Use
Casein Blocking Buffer (10×) is a 10-fold concentrated stock solution. Before use, dilute the concentrate with pure water at a ratio of 1 part concentrate to 9 parts pure water (e.g., 100 mL stock + 900 mL pure water). Mix thoroughly to prepare the 1× working solution. Prepare fresh working solution immediately before use to prevent contamination.
Protocol for Western Blot Membrane Blocking (Example):
1. Perform protein transfer and immobilization.
2. Blocking: Add an appropriate volume of 1× Casein Blocking Buffer. Incubate at 20–37 °C for a set duration (typically 15–90 minutes). Overnight blocking at 2–8 °C may yield improved results.
3. Detection: Proceed with subsequent experimental steps as required.
Note: Dilute primary and secondary antibodies with 1× Casein Blocking Buffer to achieve minimal background signals.
