Crystal Violet, also known as Gentian Violet, is a basic dye soluble in water and ethanol, which stains tissues and cell nuclei purple. It is an excellent staining agent mainly used in cytology, histology, bacteriology, and other fields.
Protocol
I. Sample Preparation
1. For Paraffin Sections
(1) Deparaffinize in xylene for 5–10 min.
(2) Replace with fresh xylene and deparaffinize for another 5–10 min; treat with absolute ethanol for 5 min.
(3) 90% ethanol for 2 min; 70% ethanol for 2 min; distilled water for 2 min.
2. For Frozen Sections
Allow sections to equilibrate to room temperature, then rinse with distilled water for 2 min.
3. For Cultured Cells
(1) Fix with 4% paraformaldehyde for ≥10 min (or follow this alternative: add 5 mL of PBS/methanol solution per 25 cm², incubate for 2 min, discard the solution; add 5 mL of fresh methanol, incubate for 10 min, discard the solution).
(2) Rinse with distilled water for 2 min.(3) Replace with fresh distilled water and rinse for another 2 min.
II. Crystal Violet Staining
Stain with 0.5% Crystal Violet Aqueous Solution for 10 min (adjust time according to staining results and requirements).
Wash thoroughly with distilled or tap water, then observe and photograph.
