Overview

IdeZ protease (IgG-specific), known as the Immunoglobulin-degrading enzyme from Streptococcus equi subspecies zooepidemicus, is a recombinant IgG-specific IdeZ protease. It is expressed via an E. coli expression system and obtained through molecular modification. This protease exhibits extremely high substrate specificity, capable of recognizing IgG from specific species (e.g., humans) and of specific types. It cleaves IgG at a specific site below the antibody hinge region, hydrolyzing IgG into intact F(ab')₂ fragments and Fc fragments (see Fig.1 for reference).

Enzyme Activity Definition: The amount of enzyme required to cleave >95% of 1 μg human IgG in a 10 μL reaction system at 37°C for 15 minutes is defined as 1 activity unit (U).

Product Applications

Suitable for structural characterization analysis of antibody drugs or antibody-fusion protein drugs. The antibody fragments prepared by enzymatic digestion can also be used to eliminate the interference of heavy chains on the detection of target proteins in Western Blot (WB) assays after immunoprecipitation, among other applications.

Operation Procedure

1. Add the required amount of IgG substrate (concentration range: 0.5~10 mg/mL) to digestion buffer or other compatible buffers.
2. Add IdeZ protease to the IgG sample, with digestion performed at a ratio of 1 U IdeZ protease per 1 μg IgG.
3. Add Reductant buffer to a final concentration of 1 mM.
4. Incubate at 37°C for 60 minutes.

Storage Conditions

1. This product can be stored stably at -20°C for 1 year. To avoid repeated freeze-thaw cycles, aliquot into small volumes for storage, and limit freeze-thaw cycles to no more than 3 times.
2. Avoid prolonged contact of Reductant buffer with air.

2. For operational safety and health protection, wear a lab coat, disposable gloves, and a mask during operation.
