PTM is the abbreviation for Pichia Trace Minerals, referring to trace elements specially formulated for Pichia pastoris. PTM1 is a trace element formula for fermentation widely applied to the PichiaPink 1, 2, 3 and 4 strain series.It is not added to seed culture medium or the initial fermentation medium at inoculation, and is mostly supplemented together with fed-batch nutrients. At present, it is also adopted in fed-batch trials for fermentation of other Pichia pastoris strains.The fermentation process of Pichia pastoris consists of four phases:1. Seed culture stage. 2.Glycerol batch phase: the period from inoculation until complete depletion of glycerol in the initial medium. This phase mainly aims to accumulate biomass and prepare for subsequent induced protein expression.3.Glycerol fed-batch phase: no protein induction is carried out in this stage, whose sole purpose is to boost biomass concentration.4. Methanol fed-batch induction phase: this phase is dedicated to inducing target protein expression.

PTM1 is sterile-filtered through a 0.22 μm filter and can be added directly for use.PTM1 Trace Element Stock Solution is designed to supplement trace elements during Pichia pastoris fermentation experiments.Fermentation protocols differ for various Pichia pastoris strains, so please follow your specific protocol accordingly.Taking the commonly used PichiaPink 1, 2, 3 and 4 strains as an example:During the glycerol fed-batch phase, start feeding 50% glycerol solution supplemented with 12 mL/L PTM1 at a feeding rate of 5–15 mL/(h·L initial fermentation volume). Continue feeding for no less than 4 hours until the wet cell weight reaches 150–220 g/L.
Precautions:
1. This PTM1 Trace Element Stock Solution has undergone 0.22 μm sterile filtration and is ready for direct use. Discontinue use immediately if bacterial contamination is detected.
2. A small amount of precipitate may form at the bottom of the solution due to its high concentration. This does not affect product performance, and the precipitate will dissolve completely once mixed into the culture medium.
