The activity of Recombinant Human Coagulation Factor II/Thrombin Protein (rp303098) was measured by the rate of free chromophore formation from Chromozym TH thrombin substrate monitored by absorbance at 405 nm. Activity ranges from 1000-5000 units/mg.
Materials:
Buffer : 50mM Tris-HCl, 0.1% BSA and 227 mM NaCl, pH 8.3
Substrate: Chromozym TH (B1008216)
Procedures:
1. Dilute Recombinant Human Coagulation Factor II/Thrombin Protein (rp303098) to 0.28 µg/mL in Assay Buffer.
2. Dilute Chromozym TH (B1008216) to 400 µM in Assay Buffer.
3. Load 50 µL of 0.28 µg/mL Recombinant Human Coagulation Factor II/Thrombin Protein (rp303098) into a plate, and start the reaction by adding 50 µL of 400 µM Substrate. Include a Substrate Blank containing 50 µL of Assay Buffer and 50 µL of Substrate.
4. Read at 405 nm (absorbance) in kinetic mode for 15 minutes.

3 μg/lane of Recombinant Human Coagulation Factor II/Thrombin Protein was resolved with SDS-PAGE under reducing (R) and non-reducing (N) conditions and visualized by Coomassie® Blue staining, showing the bands at 31.7 & 5.6 kDa under reducing conditions and 35.0 & 7.0 kDa under non-reducing conditions.
