Soil dehydrogenase activity reflects the number of active microorganisms in soil and their capability to degrade organic pollutants, and serves as an important indicator for evaluating soil biodegradability. The traditional assay method employs 2,3,5‑triphenyltetrazolium chloride (TTC) as a hydrogen acceptor; however, its reduction product formazan is water‑insoluble and requires organic solvent extraction, resulting in cumbersome operation and relatively low sensitivity. This kit provides a simple, sensitive, and rapid method for the determination of soil dehydrogenase activity. This method employs a modified water‑soluble tetrazolium salt as a hydrogen acceptor in place of TTC. The colored formazan product generated by enzymatic reduction is directly water‑soluble, eliminating the need for an extraction step. This formazan product exhibits a characteristic absorption peak at 460 nm. By measuring the absorbance of the reaction solution at 460 nm—which is directly proportional to the dehydrogenase activity—the soil dehydrogenase activity can be rapidly quantified.
