Superoxide dismutase (SOD, EC 1.15.1.1) is widely distributed in animals, plants, microorganisms, and cultured cells. It catalyzes the dismutation of superoxide anion (O₂⁻·) to H₂O₂ and O₂. SOD acts both as a scavenger of superoxide anion and a key enzyme in the generation of H₂O₂, playing an important role in the biological antioxidant system. The principle of this kit is as follows: The xanthine and xanthine oxidase reaction system generates superoxide anion (O₂⁻·), which reacts with WST-8 to form a water-soluble formazan dye with a characteristic absorbance peak at 450 nm. SOD scavenges O₂⁻·, thereby inhibiting the formation of formazan. The higher the absorbance (i.e., the more intense the yellow color), the lower the SOD activity; conversely, the lower the absorbance, the higher the SOD activity. By measuring the absorbance at 450 nm, SOD enzyme activity can be calculated.
