Overview

  Acid Phosphatase (ACP) is widely distributed in various tissues and primarily localized in the lysosomes of cells, thus often serving as a lysosomal marker enzyme. Extracellular ACP exists in the endoplasmic reticulum and cytoplasm. The properties of ACP vary across different animal species, with an optimal pH range of 4.5-5.5. ACP belongs to a protein family, and its molecular weight in mammals ranges from 18 kDa to 100 kDa. It is classified into two types: tartrate-sensitive ACP and fluoride-sensitive ACP. Lysosomal ACP is tartrate-sensitive, whereas ACP in red blood cells and macrophages is fluoride-sensitive.
  Plant Acid Phosphatase (ACP) Extraction Reagent is specially formulated for lysing plant tissues and extracting ACP from plant samples. This reagent is for research use only and not intended for clinical diagnosis or any other purposes.
Materials to Be Prepared by Users
1. Distilled water
2. Centrifuge tubes or test tubes, homogenizer or mortar and pestle, refrigerated centrifuge
Operating Procedures (For Reference Only)
1. Weigh an appropriate amount of plant tissue, rinse it thoroughly, and cut it into small pieces.
2. Add the Plant Acid Phosphatase (ACP) Extraction Reagent at a ratio of 1 g plant tissue: 2 ml reagent, then homogenize or grind the mixture thoroughly.
3. Allow the mixture to stand for 30 minutes, then filter it through gauze or filter paper.
4. Centrifuge the filtrate at 4000×g for 20 minutes. Collect the supernatant, measure its volume, and store it at -20°C for subsequent ACP activity assays or other applications.
Calculation
  Yield of crude enzyme solution from plant tissue (ml) = Volume of supernatant (ml)/Weight of plant tissue (g)×100%
Precautions
1. The samples to be tested must be free of phosphatase inhibitors. Repeated freeze-thaw cycles of the samples should also be avoided.
2. If the measured value of the sample exceeds the upper limit of the standard curve, dilute the sample with Plant Acid Phosphatase (ACP) Extraction Reagent and perform the assay again.
3. Use the reagent as soon as possible after opening to prevent adverse effects on subsequent experimental results.
4. For your safety and health, wear a lab coat and disposable gloves during operation.
