Overview

Streptavidin conjugates are widely used together with a conjugate of biotin for specific detection of a variety of proteins, protein motifs, nucleic acids and other molecules since streptavidin has a very high binding affinity for biotin. This Streptavidin-R-PE conjugate comprises streptavidin (as the biotin-binding protein) with RPE covalently attached (as the fluorescent label). It is commonly used as a second step reagent for indirect immunofluorescent staining, when used in conjunction with biotinylated primary antibodies. It is a very valuable tool for biotin-streptavidin-based biological assays and tests using flow cytometry, microplate reader and microarray platforms. This Streptavidin-R-PE product has been tested by immunofluorescent staining with flow cytometric analysis and Multiplex bead-based assay.

Multiplex bead-based assay and High Performance assays use color-coded polystyrene or superparamagnetic beads coated with target-specific antibodies. When these pre-coated beads are incubated with the sample mixture, the target analytes are captured by the beads. These captured analytes are then detected by a cocktail of biotinylated detection antibodies. Following the addition of a streptavidin-PE reporter, chemiluminescence is generated and detected.

Flow cytometry analysis of CD45 expression by Jurkat cells (red) compared to an isotype control (blue). Cells were stained with Recombinant CD45 Antibody (Biotin) (Ab213118) at 0.5μg/mL for 1 hour at 4°C. Then incubated with Recombinant Core Streptavidin Protein (PE) (rp230628) at 0.3μg/mL for 30 minutes at 4°C in the dark. Blue - Mouse IgG (Biotin) (Ab180306). Black - Unlabelled control, cells without incubation with primary antibody.
