This product is suitable for Western Blot experiments of chemiluminescence and fluorescence detection, and can be reused on nitrocellulose or PVDF membranes that have been tested. At room temperature, primary and secondary antibodies on the membrane can be removed without affecting the protein on the membrane, without the need for repeated electrophoresis and membrane transfer, saving samples and time.
This product has a mild formula, simple and fast operation, and can perform 3-5 Western Blot tests on the same membrane.
Precautions:
1. If precipitation occurs in the solution before use, it can be heated at 37 ° C for 5 minutes to promote dissolution.
2. It is recommended to first detect the target protein with low expression levels, and then treat it with regeneration solution and detect the protein with high expression levels.
3. It is not recommended to use BSA sealed imprinting membranes, as well as DAB, NBT/BCIP stained imprinting membranes.
Instructions for Use:
1. Remove the tested membrane from the machine, clean it with PBST or TBST for 10 minutes, and discard the cleaning solution.
2. The duration of elution using regeneration solution mainly depends on the protein expression level, antibody type, and concentration. If the detection signal is not strong, it can be eluted by shaking at room temperature for 30-60 minutes; If it is a sample with high expression and strong signal, 20mM DTT or 100mM β - mercaptoethanol can be added according to its usage volume, and shaken at room temperature for 30-60 minutes for elution.
3. Discard the regeneration solution and rinse 3 times with an appropriate volume of PBST or TBST, each time for 5 minutes.
4. At this point, the antibodies on the membrane have been eluted and can be resealed for the next round of testing.
