Diamine oxidase (DAO, EC 1.4.3.6) is widely distributed in animals, plants, and microorganisms. It catalyzes the oxidation of diamines to generate aldehydes, and its activity is closely associated with nucleic acid and protein synthesis. This kit employs an NADH‑coupled enzymatic method for the determination of DAO activity: DAO catalyzes the oxidation of diamine substrates to produce the corresponding aldehydes and ammonia (NH₃). The generated ammonia, in the presence of glutamate dehydrogenase (GDH), reacts with α‑ketoglutarate and reduced nicotinamide adenine dinucleotide (NADH) to form glutamate, while simultaneously oxidizing NADH to NAD⁺. By monitoring the rate of decrease in the characteristic absorbance peak of NADH at 340 nm, under substrate‑saturated conditions, the initial linear decrease in OD₃₄₀ per unit time (ΔOD₃₄₀/min) is proportional to DAO enzyme activity. By referencing a standard curve, the DAO enzyme activity in the sample can be precisely calculated.
