Overview

DBCO-modified Phycoerythrin (DBCO-PE) produced by Aladdin is a ready-to-use click chemistry conjugation reagent, prepared by covalently modifying high-purity R-Phycoerythrin with DBCO-PEG5-NHS ester. This product enables rapid and efficient copper-free SPAAC (strain-promoted azide-alkyne cycloaddition) with azide-modified proteins, antibodies, oligonucleotides, polymers, or other molecules under mild conditions.

 

Phycoerythrin (PE, R-PE) is a red-emitting fluorescent protein-chromophore complex that can be excited by the 488-nm blue, 532-nm green, or 561-nm yellow-green laser with increasing efficiency, and detected using a 586/14 nm bandpass filter. PE has an excitation peak at 565 nm and an emission peak at 578 nm. R-PE is a large molecule (~240 kDa) used for fluorescence-based detection, primarily in flow cytometry, microarray assays, ELISAs, and other applications that require high sensitivity but not photostability. DBCO (dibenzocyclooctyne) is a cyclooctyne that undergoes efficient, copper-free click chemistry with azides under mild conditions. This reaction features high selectivity, fast kinetics, and excellent biocompatibility, making it suitable for live-cell and in vivo applications. The introduction of a polyethylene glycol (PEG) spacer enhances the reagent’s solubility, stability, and biocompatibility, reduces non-specific binding, and prolongs circulation half-life. DBCO-PE combines the advantages of phycoerythrin and DBCO groups, enabling efficient, copper-free conjugation to azide-modified proteins, peptides, and oligonucleotides, and is well-suited for flow cytometry, imaging, and assay development.

This DBCO-PE product is purified by size exclusion chromatography to remove unconjugated molecules, ensuring optimal staining performance.

Usage Suggestion

Storage conditions: The lyophilized powder should be stored dry and protected from light at 2–8°C for long-term preservation. Do not freeze. After reconstitution, the solution is recommended to be stored at 4°C for no more than 1 week. Phycobiliproteins are sensitive to freeze-thaw cycles. Avoid exposure to heat and light.

Copper-Free click reaction:

1.    For azide-modified antibody labeling as an example: DBCO-PE is recommended at a concentration of 5 mg/mL, with a labeling mass ratio of IgG:PE = 1:3. Mix DBCO-PE with the azide-modified protein or antibody.

2.     Incubated for 2-4 hours at room temperature or overnight at 4°C.

3.     Depending on your application, you can use the conjugated protein immediately or purify the conjugated protein by size exclusion chromatography columns and resins.
