Glutamine (Gln), also known as 2-amino-4-carbamoylbutanoic acid, is the amide of glutamic acid. L-Glutamine is an encoded amino acid involved in protein synthesis and a non-essential amino acid for mammals, which can be synthesized from glucose in vivo. It has a CAS number of 56-85-9 and a molecular weight of 146.15. Glutamine appears as white crystals or crystalline powder, soluble in water (25 mg/mL), but insoluble in methanol, ethanol, ether, benzene, acetone, chloroform and ethyl acetate. It is odorless with a slight sweet taste. It is unstable in neutral solutions and easily decomposes into glutaminol or undergoes propionylation to form pyrrolidonecarboxylic acid in alcohols, alkalis or hot water.
L-Glutamine Solution (0.1 mol/L) is composed of L-glutamine, 0.9% sodium chloride and water, and is sterilized by filtration. It can be widely used as an additive in culture media for mammalian cell culture. For research use only. Not intended for clinical diagnosis or other purposes.
Operating Procedures (For Reference Only)
Operate according to the specific requirements of the experiment. The general working concentration is 2 mmol/L, which can be prepared by mixing culture medium: L-Glutamine Solution (0.1 mol/L) at a ratio of 50:1.
Precautions
1. Avoid repeated freezing and thawing, and ensure aseptic operation.
2. With a water solubility of 25 mg/mL, the 0.1 mol/L L-Glutamine Solution may present white clumps when stored at low temperature, and some insoluble substances may remain after thawing at room temperature. The solution should be incubated in warm water at 50-60°C to facilitate complete dissolution, then aliquoted under sterile conditions for later use. The aliquoted solution should be stored at -20°C promptly.
3. Glutamine is unstable in solution and can decompose by 50% after 1 week of storage at 4°C. Therefore, it should be added to the culture medium immediately before use.
4. If the culture medium supplemented with glutamine is stored in a 4°C refrigerator for more than 2 weeks, the original amount of glutamine should be replenished.
5. Use the reagent as soon as possible after opening to prevent adverse effects on subsequent experimental results.
6. For your safety and health, wear a lab coat and disposable gloves during operation.
