The LBA4404 strain has an Ach5-type background, with a screening marker—the rifampicin resistance gene Rif—in its nuclear genome. To facilitate transformation operations, this strain carries an octopine-type Ti plasmid pAL4404 that lacks self-transport functionality. This plasmid contains vir genes (which are essential components for the insertion of T-DNA into the plant genome; although the T-DNA transfer function of the pAL4404 plasmid itself is disrupted, it can assist in the smooth transfer of T-DNA from the introduced binary vector). The pAL4404-type Ti plasmid contains a screening marker: Strep, which endows the LBA4404 strain with streptomycin resistance and is suitable for transgenic operations in plants such as tobacco, tomato, etc.
