Overview

This product is a ready-to-use, sterile-filtered concentrate of Phytohemagglutinin-P (PHA-P). PHA-P is derived from red kidney beans (Phaseolus vulgaris) and represents the protein form of PHA prior to the separation of subunits, containing a mixture of glycoproteins with mitogenic and agglutinating activities. Provided as a 500X concentrated solution, it is designed for convenient direct dilution in cell culture to efficiently induce mitosis and proliferation of human and mouse peripheral blood mononuclear cells (PBMCs).

Usage Protocol

500X concentrate. The recommended final stimulation concentration ranges from 1-10 μg/mL, depending on cell type and experimental system; optimization via pilot experiments is advised.

Primary Applications

  • Nonspecific activation and proliferation assays of T lymphocytes.
  • Culture of peripheral blood mononuclear cells (PBMCs) and induction of cytokine secretion.
  • Lymphocyte transformation tests.
  • Positive control stimulant in immune cell functional studies (e.g., ELISPOT, flow cytometric cytokine detection).

Human peripheral blood lymphocytes were stimulated with Phytohemagglutinin-P (PHA-P) Solution (500X) (P1508369) for 3 days. The cells were then stained with Recombinant IL-2 Receptor alpha/CD25 Antibody (APC) (Ab230835) (red) compared to an isotype control (blue). Blue - Mouse IgG (APC) (Ab180309). Black - Unlabelled control, cells without incubation with primary antibody. Unstimulated PBMCs were used as control (left panel).
