LumiDye™ Streptavidin Azide (N₃-SA) is a ready-to-use click chemistry conjugation reagent, produced by covalent modification of high-purity streptavidin with azide groups. This product enables rapid and efficient covalent conjugation with terminal alkyne- or DBCO-modified antibodies, oligonucleotides, polymers, or other molecules under mild conditions.
Streptavidin possesses distinct biological properties: each molecule consists of four subunits capable of binding four biotin molecules with extremely high affinity, exhibiting a binding constant as high as 10¹⁵ M⁻¹, making it widely applicable in areas such as biological detection, separation, and targeted delivery. The azide group is chemically reactive and participates in click chemistry reactions, such as the classic copper-catalyzed azide-alkyne cycloaddition (CuAAC), which efficiently reacts with alkyne-containing biomolecules under mild conditions to form a stable triazole ring structure. N₃-SA combines the advantages of streptavidin and azide groups, enabling specific labeling and conjugation of biomolecules—first through streptavidin’s selective binding to biotinylated molecules, followed by the click reaction between the azide and alkyne-functionalized molecules. This allows for precise connection of various biomolecules, providing a convenient tool for biological research, with the entire process conducted under relatively mild conditions that minimally affect the original activity of biomolecules.
Usage Suggestion
Storage conditions: The lyophilized powder should be stored dry and protected from light at 2–8°C for long-term preservation. After reconstitution, the solution is recommended to be stored at 4°C for no more than 1 week, or aliquoted and frozen at –20°C.
Recommended click reaction system: Azide-alkyne reactions are typically carried out in the presence of CuSO₄, a ligand (e.g., THPTA or TBTA), and a reducing agent (e.g., ascorbic acid).
Typical reaction conditions: 0.1–1 mg/mL azide-modified streptavidin (N₃-SA), 2–5 fold molar excess of alkyne-target molecule, reaction at room temperature for 30–60 minutes.
For intracellular labeling, it is recommended to use a copper chelator or replace with copper-free click chemistry (SPAAC).
