Overview
α-1,3-N-Acetylgalactosaminyltransferase (BgtA), EC 2.4.1.40, is derived from Helicobacter mustelae. This enzyme is used for the synthesis of blood group A antigens and transfers N‑acetylgalactosamine (GalNAc) to the non‑reducing end of glycan chains with the Fucα1‑2Galβ1‑4Glcβ‑ motif. This product is recombinantly expressed in Escherichia coli with high activity.
Add substrates to the reaction system, along with 5 mM MgCl₂. Appropriate buffer salts such as Tris‑HCl may be supplemented to adjust the pH to 7.5, then add an appropriate amount of BgtA according to reaction requirements.
After mixing, incubate the sample at 37 °C for 2–4 hours or overnight.
Note: For scaled‑up reactions, incubation time or enzyme dosage shall be adjusted according to practical conditions; low‑speed shaking can be applied for homogenization.
