Carbon dioxide (CO₂) is one of the important products of energy metabolism in living organisms. Its quantification is of broad significance in respiratory metabolism research and clinical biochemical testing. This kit employs a PEPC-MDH coupled enzymatic method for the determination of CO₂ content in samples. In the presence of phosphoenolpyruvate carboxylase (PEPC) and Mg²⁺, phosphoenolpyruvate (PEP) reacts with HCO₃⁻ to generate oxaloacetate and phosphate. The oxaloacetate is then reduced to malate by NADH under the catalysis of malate dehydrogenase (MDH), with concomitant oxidation of NADH to NAD⁺. In this coupled reaction system, the rate of NADH consumption is directly proportional to the CO₂ content in the sample. By monitoring the rate of decrease in the characteristic absorbance peak of NADH at 340 nm and referencing a standard curve, the CO₂ content in the sample can be precisely calculated.
