Overview
Diamine oxidase (DAO, EC 1.4.3.6) is widely distributed in animals (including intestinal mucosa, lung, liver, kidney, etc.), plants, and microorganisms. It catalyzes the oxidation of polyamines to aldehydes. Its activity is closely associated with nucleic acid and protein synthesis and can reflect the integrity and degree of damage of the intestinal mechanical barrier.
The assay principle of this kit is as follows: DAO catalyzes the oxidation of cadaverine to generate aldehydes and hydrogen peroxide (H₂O₂). In the presence of excess horseradish peroxidase (HRP), H₂O₂ oxidizes o-dianisidine to its oxidized form, which exhibits a characteristic absorbance peak at 460 nm. By measuring the rate of increase in absorbance at this wavelength, DAO enzyme activity can be calculated.
