The EHA101 strain is derived from the C58 background, with a screening marker—the rifampicin resistance gene Rif—present in its nuclear genome. For facilitating transformation operations, this strain carries a nopaline-type Ti plasmid pEHA101 (pTiBo542ΔT-DNA) that lacks self-transport functionality. This plasmid contains vir genes, which are essential components for the insertion of T-DNA into the plant genome. Although the T-DNA transfer function of the pEHA101 (pTiBo542ΔT-DNA) plasmid itself is disrupted, it can assist in the smooth transfer of T-DNA from the introduced binary vector. The pEHA101 (pTiBo542ΔT-DNA) type Ti plasmid contains a screening marker: Kan, which endows the EHA101 strain with kanamycin resistance. It is suitable for transgenic operations in plants such as maize, rice, and tobacco.
