The active ingredient of this product is palmitic acid at a concentration of 6 mM. It is a sterile solution filtered through a 0.22 μm membrane and can be directly mixed with complete culture medium for experimental use. The solvent adopted in this product contains neither sodium hydroxide nor organic solvents, thus exerting no cytotoxicity to cells. This supplement can be applied to induce cellular lipotoxicity, endoplasmic reticulum stress, oxidative stress, mitochondrial damage, cell apoptosis and other related cellular phenotypes.
Instructions for Use
1.The working concentration of this product varies depending on cell types and experimental objectives. Generally, dilute it with cell culture medium at a volume ratio ranging from 1:120 to 1:10.
2.For first-time use, treat cells with a concentration gradient to identify the optimal concentration that yields the desired effect for your specific cell line.
3.When cells reach 80–100% confluence, replace with fresh culture medium. Add this product dropwise at the appropriate dilution ratio. Add an equal volume of vehicle control to the control group, mix gently, and place the cells back into the incubator for further incubation.
4.Harvest cells 20–48 hours after treatment for subsequent analysis.
Precautions
Upon receipt, aliquot the product for long-term storage at -20 °C; short-term storage at 2–8 °C is acceptable for up to 3 months.
The solvent containing BSA may interfere with absorbance readings. When measuring cytotoxicity via CCK-8 (Aladdin Cat. No. C266180), aspirate the mixture of high-fat cell supplement and culture medium, wash the cells 2–3 times with PBS, then add CCK-8 working solution.
