Nitrogen is one of the essential elements for living organisms. The nitrogen cycle in nature involves multiple transformation processes: atmospheric nitrogen (N₂) is fixed into ammonia‑nitrogen (NH₄⁺‑N) by nitrogen‑fixing microorganisms and plant‑microbe symbionts, which is then converted to nitrate‑nitrogen (NO₃⁻‑N) through the action of nitrifying microorganisms, and subsequently assimilated by plants or microorganisms into organic nitrogenous compounds. Therefore, the ammonia‑nitrogen content in plant tissues can reflect the degree of stress they are experiencing. The assay principle of this kit is based on the ninhydrin color reaction: α‑amino acids are heated with ninhydrin hydrate, undergoing oxidative deamination to yield the corresponding α‑keto acids, which are decarboxylated to form aldehydes, while ninhydrin hydrate is reduced. In a weakly acidic environment, the reduced ninhydrin, ammonia, and another molecule of ninhydrin hydrate condense to form a blue‑purple product that exhibits a characteristic absorption peak at 580 nm. By measuring the absorbance, the amino acid content can be quantitatively analyzed.
