Overview
The anthocyanin content is determined using the pH differential method. At pH 1.0, anthocyanins exhibit a characteristic absorption peak at 530 nm. At pH 4.5, anthocyanins are converted to the colorless chalcone form, which shows no absorption at 530 nm. By taking advantage of this property, the absorbance values at 530 nm and 700 nm are measured under the two pH conditions. The anthocyanin content in plant samples can then be quantitatively determined.
