Overview

Soil polysaccharides are an important component of soil organic matter research. Numerous studies have demonstrated that soil polysaccharides play a crucial role in promoting the formation of water‑stable aggregates, enhancing soil structural stability, improving soil erosion resistance, and increasing soil water and nutrient retention capacity. Therefore, the determination of soil polysaccharide content is of significant research importance. This kit employs the phenol‑sulfuric acid colorimetric method for the determination of soil polysaccharide content. The principle is as follows: Polysaccharides in soil samples are hydrolyzed to monosaccharides under the action of concentrated sulfuric acid. The monosaccharides are rapidly dehydrated to generate furfural derivatives, which then condense with phenol to form an orange‑yellow compound. This product is stable in color and exhibits a characteristic absorption peak at 488 nm, with its absorbance showing a linear positive correlation with polysaccharide content within a certain concentration range. By measuring the absorbance at this wavelength and referring to a standard curve, the soil polysaccharide content in the sample can be accurately quantified.
