α-Ketoglutarate (α-KG) is an important intermediate metabolite in the tricarboxylic acid (TCA) cycle, positioned after isocitrate and before succinyl-CoA. It links carbon and nitrogen metabolism, directly participating in oxidative energy production and the biosynthesis of various substances, thus playing a vital role in maintaining normal physiological functions. This kit provides a rapid and sensitive method for the determination of α-KG: Under the catalysis of alanine aminotransferase (ALT), α-KG is converted to glutamate. The glutamate is then further reacted by glutamate dehydrogenase (GDH) to generate NADH. The generated NADH reacts with a chromogenic reagent to produce a yellow product with a characteristic absorbance peak at 450 nm. By measuring the absorbance at this wavelength, the α-KG content in the sample can be precisely calculated.
