Overview

Ac-LEHD-AMC is a small-molecule fluorescent substrate widely used in the detection of Caspase 9 activity. This substrate contains a specific peptide sequence (LEHD), which is a targeted substrate for Caspase 9. It can be cleaved under the catalysis of Caspase 9 to release AMC (7-amino-4-methylcoumarin), producing blue fluorescence. The maximum absorption wavelength of Ac-LEHD-AMC is usually 360 nm, and the emission wavelength is 450 nm, making it very suitable for fluorescence detection. The compound usually exists in the form of a white solid or powder, and is easily soluble in DMSO and water.
Ac-LEHD-AMC is a substrate made by linking the Caspase 9 substrate peptide (LEHD) to the fluorophore AMC. Caspase 9 is a member of the cysteine-aspartic protease family and is involved in the endogenous pathway of cell apoptosis, especially playing an important role in the death receptor pathway and mitochondrial pathway. LEHD is a specific recognition sequence for Caspase 9 and can be cleaved by Caspase 9. The release of the AMC fluorophore makes this substrate an ideal tool for fluorescent detection of Caspase 9 activity.

Structural features of Ac-LEHD-AMC include:
