Immunohistochemical analysis of Smoothened staining in human breast cancer formalin fixed paraffin embedded tissue section. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0). The section was then incubated with Smoothened Antibody (Ab128394) at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.

IF analysis of Smoothened Antibody (Ab128394) in HeLa cells. The cells were fixed with 4% PFA for 10 minutes, and blocked with 5% BSA for 1 hour at room temperature. Cells were stained with Smoothened Antibody (Ab128394) at 1/200 dilution in blocking buffer overnight at 4°C, and then incubated with Goat Anti-Rabbit IgG H&L (AF647) (Ab181958) at a dilution of 1/1000 for 1 hour at room temperature in the dark (red). Cells were counterstained with DAPI (blue).
