Overview

Alkaline protease is a proteolytic enzyme produced from Bacillus licheniformis strains bred by protoplast mutagenesis, via submerged fermentation, extraction and purification. It is widely used in leather processing, silk degumming, food, medical, brewing and other industries.

Principle

The active component of alkaline protease is a serine endo-alkaline protease, which hydrolyzes peptide bonds in protein molecules to produce polypeptides or amino acids. It can also catalyze the synthesis of polypeptides in organic solvents.

Product Characteristics
1. Temperature range: Effective at 20–60°C, optimum at 35–45°C.
2. pH range: Effective at pH 6–11, optimum at pH 9.5–10.5.
3. Effect of metal ions on enzyme activity: Ca²⁺, Mn²⁺, Mg²⁺, Zn²⁺ and Fe²⁺ show activating effects. The enzyme is insensitive to metal chelator EDTA, heavy metals and thiol reagents. Reagents acting on serine residues (such as sulfonyl halides) will inactivate the enzyme.
4. Product standard: Complies with GB/T 23527-2009, the national standard of the People's Republic of China.
5. This product is industrial grade. The excipients contain anhydrous sodium sulfate and other components, which may result in incomplete dissolution.
Application Methods
1. Leather processing: Simplifies process, shortens cycle, improves quality, increases yield and reduces cost. Soaking: 0.01–0.05% (based on raw material weight; enzyme activity 200,000 U/g, same below), 20–25°C for 12–20 h. Bating: 0.025–0.1%, 35–38°C for 3–6 h. Depilation: 0.05–0.15%, 20–35°C for 12–20 h.pH for all above: 9–11.
2. Silk degumming: Minimizes damage to fibroin, avoids fuzzing and improves softness. After pretreatment, dosage 0.4–1.2%, pH 9–11, 40–50°C for 30–60 min.
3. Chondroitin production: Improves yield and purity. After alkaline extraction, dosage 0.1–0.3%, pH 8–10, 40–50°C for 4–8 h.
4. Heparin sodium production: Enhances molecular uniformity and product purity. After salt hydrolysis, dosage 1–2 g per small intestine, 45–60°C, pH 9–11 for 4–6 h.
5. Bioactive peptide & protein processing: Improves nutritional value and digestibility. Solid-liquid ratio 1:3–9, pH 8–10, 50–60°C, dosage 0.25–0.75%, hydrolysis for 2–6 h.
6. Detergent applications: Granulated and coated enzyme used in laundry powder and detergents greatly improves removal of protein-based stains (blood, sweat, milk). Dosage: 0.1–0.5% based on detergent weight.
Application Notes

Alkaline protease is a natural enzyme preparation and harmless to humans. However, it may cause allergies in some sensitive individuals, and prolonged contact may irritate skin, eyes and mucous membranes. Protective equipment such as masks and goggles is recommended during handling.

Storage

This product is biologically active; high temperature, strong acids and strong alkalis cause inactivation. During transportation and storage, liquid enzymes shall be protected from direct sunlight, and solid enzymes from sun exposure and rain. Store in a clean, cool and dry warehouse.
