Overview

Cholesterol, also known as cholesterin, is a derivative of cyclopentanoperhydrophenanthrene widely distributed in animals. It is most abundant in the brain and nervous tissue, with relatively high levels also found in the kidneys, spleen, skin, liver, and bile. The enzymatic method for determining Total Cholesterol (TC) is a common procedure in biochemical testing. In blood, approximately one-third of cholesterol exists in the free form, while the remaining two-thirds is esterified with fatty acids. In the assay, cholesterol esterase (CEH) hydrolyzes cholesterol esters into free cholesterol. The free cholesterol is then oxidized by cholesterol oxidase (COD) to form cholestenone, with the concomitant production of hydrogen peroxide. Subsequently, in a reaction catalyzed by peroxidase (POD), the hydrogen peroxide facilitates the reaction of 4-aminoantipyrine with phenol (collectively termed PAP) to generate a red-colored quinoneimine dye (the Trinder reaction). This product is quantified colorimetrically using a spectrophotometer at a wavelength between 500 and 520 nm. This assay kit is primarily designed for the quantitative determination of total cholesterol content in samples such as human or animal serum, plasma, cerebrospinal fluid, cells, and tissues. It is intended for research use only and is not suitable for clinical diagnosis or other purposes.
