Overview

α-Galactosidase (α-GAL, EC 3.2.1.22) is widely present in animals, plants, microorganisms, and cultured cells. It specifically catalyzes the hydrolysis of α-galactosidic bonds and is primarily involved in the degradation of galactosides such as raffinose, stachyose, melibiose, and galactomannan. α-GAL is crucial for seed germination in plants. During the early stages of germination, the D-galactose produced by its catalysis is rapidly converted and consumed via the glycolytic pathway, providing the initial energy source for germination. In later stages, it mainly participates in the hydrolysis of cell wall storage polysaccharides. α-GAL catalyzes the hydrolysis of p-nitrophenyl-α-D-galactopyranoside to generate p-nitrophenol, which exhibits a maximum absorption peak at 400 nm. The activity of α-GAL is calculated by measuring the rate of increase in absorbance at this wavelength.
