Overview
Alanine aminotransferase (ALT/GPT) is widely present in animals, plants, microorganisms, and cultured cells. It catalyzes a reversible transamination reaction and serves as an important enzyme in amino acid metabolism. The serum ALT activity level is a crucial biochemical indicator of liver damage, reflecting the severity of hepatic injury. Additionally, ALT is found in the highest concentration in myocardial cells and is commonly used in clinical settings as an auxiliary test for myocardial infarction and myocarditis.
This kit provides a simple method for detecting the activity level of alanine aminotransferase (ALT/GPT) in biological samples such as serum (plasma), animal and plant tissues, cells, and bacteria. Under conditions of 37°C and pH 7.4, ALT/GPT catalyzes the transamination reaction between α-ketoglutarate and alanine, producing glutamate and pyruvate. The pyruvate generated reacts with 2,4-dinitrophenylhydrazine to form pyruvate phenylhydrazone, which develops a brownish-red color under alkaline conditions. The enzyme activity of ALT/GPT can be calculated by measuring the absorbance at 505 nm.
