Overview

Alkaline phosphatase (ALP or AKP) is a type of phosphomonoesterase widely distributed in mammalian tissues, with an optimal pH range of 9.2–9.8 for its activity. This enzyme is primarily located at sites of active substance exchange (such as the cell membrane), including the brush border of intestinal epithelial cells and renal proximal convoluted tubules, the stereocilia of epididymal epithelium, the capillary bile duct membranes of the liver, and the endothelium of arterioles and arterial capillaries. It is also found in the endoplasmic reticulum, Golgi apparatus, pinocytotic vesicles, lysosomes of intestinal epithelial cells, specific granules of neutrophils, and the cell membrane of smooth muscle cells. This kit employs the disodium phenyl phosphate colorimetric method. The detection principle is as follows: under alkaline conditions, disodium phenyl phosphate is hydrolyzed by alkaline phosphatase to produce free phenol and phosphate. In an alkaline environment, phenol reacts with 4-aminoantipyrine and is oxidized to form a red quinone derivative, exhibiting varying shades of red. The intensity of the red color is proportional to the alkaline phosphatase activity—darker red indicates higher enzymatic activity, while lighter red indicates lower activity. By measuring the absorbance at 510 nm using spectrophotometry and performing colorimetric analysis, the alkaline phosphatase activity level can be calculated. This product is suitable for detecting endogenous alkaline phosphatase activity in samples such as cell or tissue lysates, homogenates, plasma, and serum. It is intended for research use only and is not suitable for clinical diagnosis or other purposes.
