Digestion Termination Solution is a type of reagent independently developed by Aladdin, designed to terminate digestion when cells are digested with trypsin. Trypsin contained in trypsin digestion solution is a serine protease, while this product contains antithrombin III, a serine protease inhibitor. Therefore, it exerts an inhibitory effect on the cell digestion process mediated by trypsin. This Digestion Termination Solution has undergone sterile treatment and can be directly used in cell experiments.
Instructions for Use (Adherent Cells):
a. After removing the culture medium, gently wash the cells once with sterile PBS to eliminate residual serum.
b. Add an appropriate amount of trypsin cell digestion solution to digest the cells (typically, 1 mL of trypsin digestion solution is added to a 25 cm² cell culture flask).
c. Observe under a microscope: when the cells shrink significantly and obvious changes in cell morphology are visible to the naked eye at the bottom of the culture vessel, stop digestion. The digestion time should be between 30 seconds and 3 minutes, and should not be prolonged.
d. Immediately add an equal volume (1 mL) of Digestion Termination Solution to the trypsin. Subsequently, gently pipette to detach the cells, then properly disperse and resuspend them.
Note: Do not over-digest the cells. The optimal digestion endpoint is when the cells can just be detached by pipetting. Over-digested cells usually cannot be used for cryopreservation, as their subsequent growth performance will be poor. The process of pipetting and resuspending the cells must be appropriately gentle; otherwise, it may affect the cell survival rate during thawing.
e. Collect the cells or proceed with subsequent experiments.
Precautions:
1. Special attention should be paid during the use of the Digestion Termination Solution to avoid contamination of the solution.
2. If the amount used each time is small, it is recommended to aliquot the product before use.
3. This product is intended solely for scientific research by professional personnel. It shall not be used for clinical diagnosis or treatment, nor for food or pharmaceutical purposes, and shall not be stored in ordinary residential premises.
4. For your safety and health, please wear a lab coat and disposable gloves during operation.
