Overview

  Fixative solutions are mainly classified into aldehyde fixatives, mercuric fixatives, alcoholic fixatives, oxidizing fixatives, picrate fixatives, etc. Among the commonly used ones are formalin from aldehydes and ethanol from alcohols.

  Clarke Fixative Solution, also known as Clarke’s solution, is mainly composed of ethanol and acetic acid. It provides favorable histological morphology for HE staining after fixation. Its advantage is that it can preserve nucleic acids. Its disadvantages are as follows:

  It tends to remove lipids, so it is not suitable for fixation in fat staining.

  It is only suitable for short‑term fixation, and tissues should be transferred to 95% ethanol after fixation.

Procedure (For Reference Only):

1. Perform the procedure according to specific experimental requirements.

2. Generally, fixation takes only 30–60 minutes.

3. For larger tissue blocks, the fixation time may be appropriately extended, but should not exceed 24 hours. After fixation, the tissue should be transferred to 95% ethanol.

Precautions:

1. Fixation time varies with the thickness of the tissue sample. Proper tissue selection facilitates penetration of the fixative.

2. The volume of fixative should be sufficient; generally, the volume ratio of fixative to tissue block should be greater than 10:1. If the volume is inadequate, the fixative may be replaced 1–3 times during fixation.

3. Fresh tissues should be fixed promptly after excision. If immediate fixation is not possible, they should be stored in physiological saline and submitted for testing without delay.

5. This product is for research use only and strictly prohibited for other purposes.
